Die Biosynthesewege der isoprenoiden Verbindungen Phytol, Geranylgeraniol, cis-Abienol und Sterole, sowie des Alkaloids Nicotin wurden in Markierungsexperimenten (in vivo) in Chloroflexus aurantiacus, Catharanthus roseus und Nicotiana tabacum (Wildtyp, RPO und NDH Mutanten) mit 13C-markierten Vorstufen [U-13C2]Glycin, [U-13C6]D-Glucose, [1-13C1]D-Glucose und 13CO2 untersucht.C. aurantiacus nutzt für die Terpenbiosynthese ausschließlich den Mevalonatweg. Translated abstract:
The biosynthetic pathways of the terpenoids phytol, geranylgeraniol, cis-abienol, sterols, as well as the alkaloid nicotin were analysed by in vivo incorporation experiments in Chloroflexus aurantiacus, Catharanthus roseus and Nicotiana tabacum (wildtyp, RPO and NDH mutants) using [U-13C2]glycine, [U-13C6]glucose, [1-13C1]glucose and sup>13CO2 as precursors.C. aurantiacus uses the mevalonate pathway for terpenoid biosynthesis. NMR spectroscopy was used to define the ratio between the two different terpenoid biosynthetic pathways in C. roseus and N. tabacum.The amino acid biosynthesis in the mutants of N. tabacum was found to be different from the wildtyp. The labeling patterns of lysine from C. roseus and N. tabacum could not be explained by the known biosynthetic pathways.The stereochemical processes of deoxyxylulose phosphate reductoisomerase and of sepiapterin reductase were delineated with selectively 2H labeled NADPH.
