- Kurzfassung:
- The immunoproteasome represents a potential drug target for the treatment of chronic inflammations and autoimmune disorders. However, the therapeutic window of immunoproteasome inhibitors as anti-inflammatory agents depends entirely on their isoform selectivity that excludes targeting of the constitutive proteasome. In this thesis, three new concepts of selective immunoproteasome inhibition and the underlying principles have been investigated by X-ray crystallography.

- Das Immunproteasom stellt eine potentielle Zielstruktur für Wirkstoffe zur Behandlung von chronische Entzündungen und Autoimmunkrankheiten dar. Jedoch hängt das therapeutische Fenster von Immunproteasom-Inhibitoren als Entzündungshemmer vollständig von deren Isoform-Spezifität ab, die eine Inhibition des konstitutiven Proteasoms verhindert. In dieser Arbeit wurden drei neue Konzepte für die spezifische Immunproteasom-Inhibiton und ihre Grundprinzipien mittels Kristallstrukturanalyse aufgeklärt.
