Abstract:: Das molekulare Chaperon Hsp70 zeigt in Krebszellen eine atypische Membranlokalisation. Diese Beobachtung konnte für verschiedene Tumormodellzelllinien im Rahmen dieser Arbeit bestätigt und näher charakterisiert werden. Neben den Ursachen der Membranlokalisation wurden direkte Hsp70-Lipidinteraktionen untersucht. Für die in vitro und in vivo Untersuchungen wurde dafür unter anderem eine stabile Zelllinie mit C-terminal markiertem Hsp70 erzeugt und eine neue Reinigungsstrategie für humanes Hsp70 aus E. coli etabliert.
Translated abstract:: In cancer cells, the molecular chaperon Hsp70 can be found on plasma membranes. This atypical localisation could be verified and further characterized for various tumor cells. Direct lipid interactions and reasons for membrane localisation of Hsp70 were analyzed. Among others a new stable cell line expressing c-terminal tagged Hsp70 and a new protein purification strategy for human Hsp70 were established in the course of these in vivo and in vitro characterization.
