Reprogramming of T cell specificity by T cell receptor (TCR)-transfer for therapeutic application represents a promising new approach for the treatment of cancers. First clinical trials clearly demonstrate this. The goal of this work is the development of novel methods for the isolation of antigen specific TCRs for this purpose. By identification of antigen specific T cells in the naïve repertoire and direct TCR isolation from single cells this goal is achieved.

T-Zell-Rezeptor (TZR)-Transfer zur Umprogrammierung der Spezifität von T-Zellen zur therapeutische Verabreichung ist ein vielversprechendes neues Verfahren zu Behandlung von Krebserkrankungen. Erste klinische Studien belegen dies deutlich. Das Ziel dieser Arbeit ist es, neue Methoden zur Isolation antigenspezifischer TZR für diesen Zweck zu entwickeln. Durch Identifikation antigenspezifischer T-Zellen im naiven Repertoire und direkte Isolation des TZR aus Einzelzellen wird dieses Ziel erreicht.
