In der vorliegenden Arbeit wurde die Dosis-Wirkungs-Beziehung der Substanzen Etoposid, Docetaxel und Imatinib (STI571) auf die Zelllinien PC3, LNCaP und DU145 mittels XTT- Assay untersucht. Auch die Wirkung der alleinigen Bestrahlung mit 5 Gy war Gegenstand der Arbeit. Zusätzlich wurden Kombinationstherapien mit den genannten Chemotherapeutika und einer Bestrahlung an jeweils unterschiedlichen Tagen analysiert. Sowohl durch alleinige Bestrahlung als auch durch Zytostatikazugabe konnte eine Hemmung der Zellproliferation erzielt werden. In der Kombination empfiehlt sich vor allem die Bestrahlung am Tag 1 und anschließende Zugabe des Chemotherapeutikums am Tag 3. Zusammenfassend erscheint ein Therapieregime mit einer Bestrahlung vorweg und anschließender Chemotherapie sinnvoll.     «

In the present work the cytotoxic effect of the substances Etoposid, Docetaxel and Imatinib (STI571) were examined on the cell lines PC3, LNCaP and DU145 using the XTT-Assay. Also the effect of radiotherapy with 5 Gy was object of these work. In addition, different setups combined the cytotoxic agents with radiotherapy on different days were evaluated. Both, the addition of the chemotherapeutic agents and radiotherapy with 5 Gy induced a significant inhibition of cell proliferation on the three cell lines. By the combination therapy it seems to be beneficial to begin with radiotherapy on day 1 and add the cytotoxic agent on day 3. In conclusion a regime with previous radiotherapy and following chemotherapy seems to be sensible.     «
